چکیده

Oomycetes are spore-forming eukaryotic microbes responsible for infections in animal and plant species worldwide, posing a threat to natural ecosystems, biodiversity food security. Genomics transcriptomics approaches, together with host interaction studies, give promising results towards better understanding of the infection mechanisms oomycetes their general biology. Significant development progress genomic studies have been achieved over past decades but further molecular processes, gene regulations still needed. The use tools such as CRISPR/Cas RNAi helped elucidate some processes involved invasion both pathogenic oomycetes. These methods provide an opportunity accurate detailed functional analysis involving various fields genomics, epigenomics, proteomics, interactomics. Functional characterisation is essential filling knowledge gaps dynamic biological processes. However, every method has advantages limitations that should be considered before choosing best investigating particular research question. Here we review transformation systems, silencing editing techniques oomycetes, how they function, which what main disadvantages.

Tomato and Tobacco Phytoene Desaturase Gene Silencing by Virus-Induced Gene Silencing (VIGS) Technique

Background and Aims: Virus-Induced Gene Silencing (VIGS) is a virus vector technology that exploits antiviral defense mechanism. By infecting plants with recombinant viruses containing host genes inserted in the viral genome, VIGS achieves the RNA silencing process. The purpose of this study was to investigate the possibility of tomato (Lycopersicon esculentum Mill.) and tobacco (Nicotiana be...

Newer Gene Editing Technologies toward HIV Gene Therapy

Despite the great success of highly active antiretroviral therapy (HAART) in ameliorating the course of HIV infection, alternative therapeutic approaches are being pursued because of practical problems associated with life-long therapy. The eradication of HIV in the so-called "Berlin patient" who received a bone marrow transplant from a CCR5-negative donor has rekindled interest in genome engin...

Sugarcane Mosaic Virus-Based Gene Silencing in Nicotiana benthamiana

Background:Potyvirus-based virus-induced gene silencing (VIGS) is used for knocking down the expression of a target gene in numerous plant species. Sugarcane mosaic virus (SCMV) is a monopartite, positive single strand RNA virus. Objectives:pBINTRA6 vector was modified by inserting a gene segment of SCMV in place of Tobacco rattle virus (TRV) genome part 1 (TRV1 or RNA1)...

Bacterial Cellular Engineering by Genome Editing and Gene Silencing

Genome editing is an important technology for bacterial cellular engineering, which is commonly conducted by homologous recombination-based procedures, including gene knockout (disruption), knock-in (insertion), and allelic exchange. In addition, some new recombination-independent approaches have emerged that utilize catalytic RNAs, artificial nucleases, nucleic acid analogs, and peptide nuclei...

Genome-editing Technologies for Gene and Cell Therapy.

Gene therapy has historically been defined as the addition of new genes to human cells. However, the recent advent of genome-editing technologies has enabled a new paradigm in which the sequence of the human genome can be precisely manipulated to achieve a therapeutic effect. This includes the correction of mutations that cause disease, the addition of therapeutic genes to specific sites in the...
